Abstract:

Genomic factors contribute to racial and ethnic group disparities in cancer risk, development and treatment. It remains yet unclear how cancer driver genes attribute to such disparities in Urothelial Bladder Carcinoma (BLCA). In this report, we sequenced exomes of 17 Chinese BLCA patients. Combining our data with over 400 BLCA somatic mutations downloaded from TCGA and ICGC, we made cross analysis of cancer driver gene among individuals and racial groups. We found two cancer genes SMAD4 and TRPM3 that are more linked to bladder cancer in Chinese population. BLCA somatic mutational heterogeneity is pervasive in all ethnic groups, which is in line with most other type of cancers. However, given the over 400 highly personalized gene mutation profiles, we found significant discordance between Asian and Caucasian populations in their BLCA driver genes. Using gene mutual exclusivity, we concluded that the Ras-Raf-MAPK pathway is related to this racial-scale driver gene discordance. Asian bladder cancer patients have a higher chance to have mutations in the Ras-Raf-MAPK pathway. We also suggest, as cancer patient cohort grows, it is feasible and important to focus on biological relationship between gene clusters or pathways, instead of single genes, in the study of mechanisms leading to the birth and development of cancer.
